Isang baytang na naman ng tagumpay ang inakyat ni Charice sa pag-ere ng pilot ng second season ng hit musical series na Glee. Sa episode kasing ito pormal na siyang pinakilala bilang si Sunshine Corazon isang Filipino exchange student. Nakipag-mash up agad siya sa bidang si Rachel sa pamamagitan ng kantang “Telephone" ni Lady Gaga. At tila hindi pa sapat ang pagpapasikat na ito ni Charice dahil solo rin siyang bumirit ng 'Listen' mula sa musical na Dream Girls bilang kanyang audition piece sa New Directions Glee Club.
